Question for Jamacia Stock Exchange

When you buy a stock and it makes gains, why does my portfolio value decrease when I sell it?
Is there a tax or penalty for selling stock? Does the penalty only occur when all of that particular stock is sold or does it occur when any amount of that stock is sold at all
